Scientific Background:

N-terminal 6xHis Thioredoxin (Trx) tagged recombinant is a redox protein of approximately 12 kDa. Its major domain is conserved in several members of the Trx family and contains a conserved Cys-Gly-Pro-Cys catalytic site. It is ubiquitous and found in many organisms, from bacteria to mammals. Trx has been shown to function in cell proliferation, redox signalling, and inhibition of apoptosis.

Trx is reduced by the NADPH-dependent flavoenzyme thioredoxin reductase, and in turn, reduces other proteins such as protein disulfide isomerase (PDI) via cysteine thiol-disulfide exchange. Trx has been reported to selectively activate the DNA binding of various transcription factors such as p53, NFkB, AP1, and the glucocorticoid receptor, resulting in altered gene transcription. Reduced Trx can provide reducing equivalents for ribonucleotide reductase, the essential enzyme for DNA synthesis, and thioredoxin peroxidase. As a cofactor for peroxiredoxins, Trx plays a direct role in the reduction of oxidative species such as H2O2.

Other notes

On occasion, small volumes of 6xHis-Trx-tag N-terminal Negative Control vials may become trapped in the product vial seal during shipping and storage. If necessary, briefly spin the vial in a tabletop centrifuge to dislodge any liquid in the vial cap. Certain products may require dry ice shipping and an additional dry ice fee may apply.
